When you start a print, should you have any filament loaded, or should it all be the 1" behind the splitter?

When you start a print, should you have any filament loaded, or should it all be the 1" behind the splitter?
That's what I thought, when I went to do the first test print though (4 cubes) it reversed out the filament. Using the K1 profile, so not sure what happened but will try again when I'm able.
